| dc.description.abstract | El objetivo general del presente trabajo era extraer pectina a partir de la pulpa de café utilizando el método convencional de hidrólisis ácida para aprovechar este residuo. Se utilizó pulpa de café cosechada de variedad arábica de región Oriente de Guatemala. La hidrólisis se realizó en medio ácido a través de la adición de ácido clorhídrico 0.5M a 90ºC durante 1h. Posteriormente se llevó a cabo una precipitación utilizando etanol al 99.9% de pureza en una relación volumen-volumen de 1:1 con respecto al agua destilada añadida en la hidrólisis. Por último, se purificó la misma mediante los procesos de filtración, centrifugación y secado. La pectina extraída fue caracterizada por medio de la determinación del grado de esterificación (GE), contenido de ácido anhidrourónico (AUA) y contenido de ceniza. El primero se obtuvo mediante una valoración de la pectina antes y después de una desterificación en un medio con NaOH 0.1N. El segundo se obtuvo a través de una relación directa entre GE y el contenido de metoxilo el cual es un parámetro obtenido a través de otra titulación. El contenido de ceniza se determinó gracias a la diferencia de masa antes y después de un proceso de calentamiento a 400ºC por 4h. La pulpa utilizada también fue caracterizada mediante la determinación de su densidad de bulto, contenido de humedad y contenido de carbohidratos totales. La primera se calculó a través del volumen que ocupada una masa específica de pulpa arreglada naturalmente. El contenido de humedad se obtuvo con la diferencia de masa antes y después de un proceso de secado a 105ºC durante 5h. Finalmente, el contenido de carbohidratos totales se determinó a través de un método cuantitativo de la reacción de Benedict donde primero fue necesario llevar a cabo una hidrólisis con ácido sulfúrico. Se determinó que la pulpa de café utilizada para la obtención de pectina tiene una humedad promedio del 74.42 ± 0.07 %, una densidad de bulto promedio de 346.89 ± 2.08 kg/m³, y un contenido de carbohidratos promedio de 47.83 ± 2.14% dentro de los rangos típicos de esta materia prima. Se encontró que a valores más bajos de pH se obtiene un mayor rendimiento de extracción de pectina. Además, con respecto a la pectina obtenida, el contenido de ácido anhidrourónico (31.07 ± 5.79%) y grado de esterificación (49.92 ± 3.32%) ambos están fuera de los rangos comunes de una pectina comercial lo cual se pudo deber a impurezas dentro de la pectina, mientras que el contenido de ceniza (3.33 ± 0.01%) está dentro del rango de una pectina comercial. Asimismo, la pectina se clasifica como una pectina de bajo metoxilo y cumple con los principales grupos característicos presentes en la estructura molecular de la pectina en un análisis del espectro IR. Se recomienda la realización de un análisis económico integral del proceso de extracción y purificación de pectina, con un enfoque en la rentabilidad. También sería valioso comparar diferentes métodos de extracción y purificación para maximizar el rendimiento. Explorar las aplicaciones específicas de la pectina obtenida, como pruebas de gelificación y solubilidad, podría permitir delimitar las aplicaciones de esta. Finalmente, se propone considerar el escalamiento de este proceso para su implementación en un ingenio cafetalero, junto con el diseño y dimensionamiento de los equipos necesarios, con el objetivo de lograr una relación costo-beneficio óptima. | spa |
| dc.description.abstract | The primary objective of this study was to extract pectin from coffee pulp using the conventional acid hydrolysis method in order to valorize this agricultural by-product. Coffee pulp from the Arabica variety harvested in the eastern region of Guatemala was used as the raw material. Hydrolysis was carried out in an acidic medium by adding 0.5 M hydrochloric acid at 90°C for 1 hour. The pectin was subsequently precipitated using 99.9% ethanol at a 1:1 (v/v) ratio with the distilled water added during hydrolysis. Finally, the extracted pectin was purified through filtration, centrifugation, and drying. The extracted pectin was characterized by determining its degree of esterification (DE), anhydrouronic acid (AUA) content, and ash content. The degree of esterification was determined by titrating the pectin before and after de-esterification in a 0.1 N sodium hydroxide (NaOH) solution. The anhydrouronic acid content was calculated from the relationship between the degree of esterification and the methoxyl content, the latter being obtained through an additional titration. Ash content was determined from the difference in mass before and after heating the sample at 400°C for 4 hours. The coffee pulp was also characterized by measuring its bulk density, moisture content, and total carbohydrate content. Bulk density was calculated from the volume occupied by a known mass of naturally packed coffee pulp. Moisture content was determined from the difference in mass before and after drying at 105°C for 5 hours. Total carbohydrate content was quantified using the Benedict reaction, following an initial hydrolysis with sulfuric acid. The results showed that the coffee pulp used for pectin extraction had an average moisture content of 74.42 ± 0.07%, an average bulk density of 346.89 ± 2.08 kg/m³, and an average total carbohydrate content of 47.83 ± 2.14%, all of which fall within the typical ranges reported for this raw material. It was also found that lower pH values resulted in higher pectin extraction yields. Regarding the extracted pectin, the anhydrouronic acid content (31.07 ± 5.79%) and degree of esterification (49.92 ± 3.32%) were both outside the typical ranges for commercial pectin, which may be attributed to impurities present in the extracted product. In contrast, the ash content (3.33 ± 0.01%) fell within the range expected for commercial pectin. Furthermore, the extracted pectin was classified as low-methoxyl pectin, and infrared (IR) spectroscopy confirmed the presence of the principal functional groups characteristic of the molecular structure of pectin. An economic assessment of the extraction and purification process is recommended, with particular emphasis on evaluating its profitability. It would also be valuable to compare different extraction and purification methods to maximize pectin yield. Investigating the specific applications of the extracted pectin, including gelation and solubility tests, could help define its most suitable industrial uses. Finally, scaling up the process for implementation in a coffee processing facility is recommended, together with the design and sizing of the required equipment, in order to achieve an optimal cost–benefit ratio. | eng |
